Purpose
TodescribeaseptictechniquesusedbyCellCulturespecialistsinhandlingproductsfromand/ormammaliancells.
Safety
Protecteyes,mucousmembranes,opencutsandwoundsfromcontactwithbiohazardmaterial.Usegloves,goggles,mask,andprotectiveclothingasnecessary.
Equipment
LaminarfloworBIOLOGicalsafetyhoodasappropriatetothehazardousnatureoftheproject.
Materials
Wipes,lint-free.
Disinfectant,orquaternaryammonium.
Alcohol.70%ethanol.
Pipets,sterile,ofappropriatesize.
Pipet-Aid,orequivalent.
Biohazardwastecontainer.
Procedure
Carryoutallculturingoperationisalaminarflowhood.
Disinfectallsurfacespriortousewithadisinfectantsolution.
Swabdowntheworkingsurfaceliberallywith70%ethanol.
Periodicallyspreadasolutionof70%ethanolovertheexteriorofglovestominimizecontamination.Replacethemiftorn.
Incaseofanyspill,spreadasolutionof70%alcoholandswabimmediatelywithnon-lintingwipes.
Discardglovesafteruseanddonotwearthemwhenenteringanyotherlabarea.
Bringintotheworkareaonlythoseitemsneededforaparticularprocedure.
Leaveawideclearspaceinthecenterofthehood(notjustthefrontedge)toworkon.Donotcluttertheareatopreventblockageofproperairflowandtominimizeturbulence.
Swabwith70%alcoholallglassware(mediumbottles,beakers,etc.)beforeplacingtheminsidethehood.
Arrangetheworkareatohaveeasyaccesstoallofitwithouthavingtoreachoveroneitemtogetatanother(especiallyoveranopenbottleorflask).
Usesterilewrappedpipetsanddiscardthemafteruseintoabiohazardwastecontainer.
Checkthatthewrappingofthesterilepipetisnotbrokenordamaged.
Inspectthevesselstobeused:
T-flask-MustbefreefromvisIBLecontaminationorbreakage,orlackcontaineridentification.Theplasticcoveringtheflaskmustbeintact. Bottles-Checkforcracks,expirationdates.
Spinnerflasks-Checkforcracks,expirationdates,andproperassembly.
Discardanybiohazardorcontaminatedmaterialimmediately.
Neverperformmouthpipetting.Pipettormustbeused.
Whenhandlingsterilecontainerswithcapsorlids,placethecaponitssideifitmustbelaidontheworksurface.
MakesurenottotouchthetipofthePipettetotherimofanyflaskorsterilebottle. Cleantheworkareawhenfinishedbywipingwith70%alcohol.
