Leukemiacellsfrompatientswithchroniclymphocyticleukemia(CLL)expressahighlyrestrictedimmunoglobulinheavyvariablechain(IGHV)repertoire,suggestingthatalimitedsetofantigensreactswithleukemiccells.Here,weevaluatedthereactivityofapanelofdifferentCLLrecombinantantibodies(rAbs)encodedbythemostcommonlyexpressedIGHVgeneswithapanelofselectedviralandbacterialpathogens.SixdifferentCLLrAbsencodedbyIGHV1-69orIGHV3-21,butnotaCLLrAbencodedbyIGHV4-39genes,reactedwithasingleproteinofhumancytomegalovirus(CMV).TheCMVproteinwasidentifiedasthelargestructuralphosphoproteinpUL32.Incontrast,noneoftheCLLrAbsboundtoanyotherstructureofCMV,adenovirusserotype2,SalmonellaentericaserovarTyphimurium,orofcellsusedforpropagationofthesemicroorganisms.MonoclonalantibodiesorhumanizedrAbsofirrelevantspecificitytopUL32didnotreactwithanyoftheproteinspresentinthedifferentlysates.Still,rAbsencodedbyagermlineIGHV1-6951p1allelefromCMV-seropositiveand-negativeadultsalsoreactedwithpUL32.TheobservedreactivityofmultipledifferentCLLrAbsandnaturalantibodiesfromCMV-seronegativeadultswithpUL32isconsistentwiththepropertiesofasuperantigen.
