Siglecs(SialicacidbindingIg-likeLectins)areI-type(Ig-type)lectinsthatbelongtotheIgsuperfamily.TheyarecharacterizedbyanN-terminalIg-likeV-typedomainwhichmediatessialicacidbinding,followedbyvaryingnumbersofIg-likeC2-typedomains(1‑4).ElevenhumanSiglecs(Siglec-1through11)havebeenclonedandcharacterized.Withintheseeleven,thereareatleasttwogroups,oneofwhichistermedtheCD33-relatedgroup.CD33-relatedSiglecsincludeCD33/Siglec-3andSiglec-5through11(1, 3).Todate,noSiglechasbeenshowntorecognizeanycellsurfaceligandotherthansialicacid.ThissuggeststhatinteractionswithglycanscontainingthiscarbohydrateareimportantinmediatingtheBIOLOGicalfunctionsofSiglecs.TheCDNAofhumanSiglec-6(alsoknownasOB-BP1andCD33L),encodesaputative442aminoacid(aa)proteinthatcontainsa15aasignalpeptide,a321aaextracellularregion,a21aatransmembraneregion(TM),andan85aacytoplasmictail(5,6).TheextracellularregioncontainsoneN-terminalV-typeIg-likedomainfollowedbytwoIg-likeC2-typedomains.Thecytoplasmicdomainhasoneimmunoreceptortyrosine-basedinhibitionmotif(ITIM).Atleastthreeadditionalisoformsexist,allofwhichencodeanadditional11aa’sattheN-terminus,likelyduetotheutilizationofanalternatestartsite.Twoofthethreeisoformsalsoshowsplicing.Oneisoformshowsa16aain-framedeletioninthesecondC2-likedomain,whiletheothershowsadeletionoftheTMandcytoplasmicregion,thuspotentiallygeneratingasolubleform(6‑9).Siglec-6isfoundonBcellsandinplacenta,andwouldseemtohavearestrictedspecificityforthesialylTnantigen(6,10).
