Background:
Syndecan-2,previouslyknownasfibroglycanorheparansulfateproteoglycan,isamemberofthesyndecanfamilyofType1transmembraneproteinscapableofcarryingheparansulfate(HS)andchondroitinsulfateglycosaminoglycans.Thefourvertebratesyndecansshowconservedcytoplasmicdomainsanddivergentextracellularportions(exceptforGAGattachmentsites).AmongtheSyndecans,Syndecan-2ismostsimilartoSyndecan-4(1‑3).HumanSyndecan-2issynthesizedasa201aminoacid(aa)coreproteinwithan18aasignalsequence,a126aaextracellulardomain(ECD),a25aatransmembraneregionanda32aacytoplasmictail(4).ThehumanECDofSyndecan-2containsthreeclosely-spacedconsensusSer-GlysequencesfortheattachmentofHSsidechains.Itshares76%,73%,87%,78%and63%aaidentitywiththeECDofmouse,rat,bovine, canineandchickenSyndecan-2,respectively.Thecytoplasmictailhasbothserineandtyrosinephosphorylationsites.Additionof20‑80disaccharidespersidechainaddsconsiderablytothesizeofthe22kDacoreprotein.Non-covalenthomodimerizationofSyndecan-2isdependentonthetransmembranedomain(5).Syndecan-2isexpressedincellsofmesenchymalorigin,neuronalandepithelialcells,andisthepredominantsyndecanexpressedduringembryonicdevelopment.Expressionisupregulatedinseveralcancercelllines(6).Afterinductioninmacrophagesbyinflammatorymediators,Syndecan-2selectivelybindsFGFbasic,VEGFandEGF(7).Syndecan-2expressedonhumanprimaryosteoblastsbindsGM-CSFandmayfunctionasaco‑receptor(8).ActivatedendothelialcellSyndecan-2specificallybindsIL-8andmayparticipateinpromotingneutrophilextravasationbyformingachemotacticIL-8grADIent(9).Typically,cytokine,chemokineandextracellularmatrixproteinbindingoccursthroughinteractionwithHSsidechains,buttheSyndecan-2extracellulardomaincanbindTGF-betadirectlyviaprotein-proteininteraction(10).
