Thisassayisintendedforresearchuseonly.
2.GeneralInformation Poliomyelitisisaninfectioncausedbyenterovirus,whichoccursepidemicallyworld-wide,and whichoftenleadstoparalysisanddeath.Threetypesofhuman-pathogenicpoliomyelitisvirusesare actuallyknown: Type1(Brunhilde):oftenwithseveresymptoms Type2(Lansing):withmildersymptoms Type3(Leon):rare,butwithseveresymptoms Poliovirusesmainlyproliferateinthelymphnodesoftheintestine,andareexcretedviafeces.The throatcanalsobeinfected,andthevirusesthenleavethebodyorally. Aftertheinfection,the virusesaredistributedviamonocytesintootherlymphnodes,wheretheymultiplicate.Inasecond viremicphasetheysettleinthewholeorganism,amongstothersinthecentralnervoussystem.In morethan90%oftheinfectionstheindividualdoesnotsufferanysubjectivesymptoms.Inthe remainingothercasesthereappear:unspecificillnesswithslightfever,headandthroatirritations, diarrhoea,nausea,andvomiting.Veryrarelytheclassicalparalysiswithafflictionofmusclesand cerebralnervesisseen.Thereconvalescentphasecanlastuptotwoyears,frequentlytherestay long-lastingdamages.Thereexistsnotreatment ofthedisease,onlysymptomatictherapywith acetylsalicylicacidandgymnasticsispossible.InmanycountriesoftheAsiaticareapoliomyelitis isstillendemic,butWHOhasstartedambitiousprojectstoerADIcatethedisease.InEuropethere existcaseswhichareimportedbytourists,sometimeswithlethaloutcome.Thediagnosisof poliomyelitisisperformedbydirectdetectionoftheinfectiousagentinstoolorthroatwashings duringtheincubationtimeofthevirus,orbydeterminationofantibodiesintheblood.Thelatteris usuallydonebytheneutralisationtest,wheretiterdifferencesofpairedseraagainstthethreevirus typesaremeasuredseparately.OnlyrecentlyaPoliomyelitisIgGELISAassaywasdevelopedin analogytoDiphtheriaandTetanus,whichcandetectantibodiesagainstthethreetypesofPolio simultaneously.ThisSEROlogicalresponsecanbeduetoapastillnessortoimmunityby vaccination.
3.PrincipleoftheTest TheGenWayPolioIgGantibodytestkitisbasedontheprincipleoftheenzymeimmunoassay (EIA).Polioantigenisboundonthesurfaceofthemicrotiterstrips.Dilutedsampleserumorreadyto-usestandardsarePipettedintothewellsofthemicrotiterplate.AbindingbetweentheIgG antibodiesoftheserumandtheimmobilizedPolioantigentakesplace.Afteraonehourincubation atroomtemperature,theplateisrinsedwithdilutedwashsolution,inordertoremoveunbound material.Thenready-to-useanti-human-IgGperoxidaseconjugateisaddedandincubatedfor30 minutes.Afterafurtherwashingstep,thesubstrate(TMB)solutionispipettedandincubatedfor20 minutes,inducingthedevelopmentofabluedyeinthewells.Thecolordevelopmentisterminated bytheadditionofastopsolution,whichchangesthecolorfrombluetoyellow.Theresultingdyeis measuredspectrophotometricallyatthewavelengthof450nm.TheconcentrationoftheIgG antibodiesisdirectlyproportionaltotheintensityofthecolor.
