| Description | T4 Endonuclease V (T4 pyrimidine dimer glycosylase, T4 PDG), has both DNA N-glycosylase and apurinic/apyrimidinic lyase (AP lyase) activities. This enzyme recognizes cis-syn-cyclobutane pyrimidine dimers in DNA caused by ultraviolet (UV) irradiation, and the endonucleolytic activity cleaves the phosphodiester bond at the AP site. |
| Applications |
|
| Source | Purified from an E. coli strain carrying a plasmid encoding T4 denV gene. |
| Components | T4 Endonuclease V: 10,000 units/ml in 10 mM Tris-HCl, 100 mM NaCl, 0.1 mM DTT, 0.1 mM EDTA, 50% Glycerol, pH 7.5 @ 25°C |
| Unit Definition | One unit of T4 Endonuclease V converts 1 µg of UV-irradiated plasmid DNA from covalently closed circles to nicked form in 30 minutes at 37°C. |
| Quality Control | The absence of nonspecific exo- and endonuclease activities and RNase activity is confirmed by appropriate quality test. |
| Storage Condition | -20 °C |
