Description
Persephinisadisulfide-linked,homodimeric,neurotrophicfactorstructurallyrelatedtoGDNF,artemin,andneurturin.Theseproteinsbelongtothecysteineknotfamilyofgrowthfactorsthatassumestabledimericstructures.Persephinsignalsthroughamulticomponentreceptorsystem,composedofRETandoneoffourGFRα(α1-α4)receptors.TheGFRα4wasfirstidentifiedinchicken,andwaslatershowntobethepreferentialbindingsubunitforpersephin.Persephinpromotesthesurvivalofventralmidbraindopaminergicneuronsandmotorneuronsaftersciaticnerveoxotomy,and,likeGDNF,promotesuretericbudbranching.However,incontrasttoGDNFandneurturin,persephindoesnotsupportthesurvivalofperipheralneurons.RecombinantHumanPersephinisadisulfide-linkedhomodimer,composedoftwo10.4 kDapolypeptidechains(194 totalaminoacidresidues).Eachchaincontainssevenconservedcysteineresidues,oneofwhich(Cys64)isusedforinter-chaindisulfidebridging,andtheothersareinvolvedintheintramolecularringformationknownasthecysteineknotconfiguration.ManufacturedunderlicensefromtheUniversityofWashingtonpatent#US6,716,600;US6,692,943;US6,645,937;US6,403,335;US6,232,449;US6,222,022.
