| Description | The human Transforming growth factor beta-1 (TGFb1) Kit is a sandwich ELISA. The capture antibody is a monoclonal TGFb1 antibody pre-coated onto the 96-well strip plates provided in the kit. Human test samples and standards of known TGFb1 concentration are added to these wells and allowed to complex with the bound TGFb1 antibody. A biotinylated TGFb1 polyclonal antibody is then added. This detection antibody binds to the antigen thus completing the sandwich. After washing, an enzyme Avidin-Biotin-Peroxidase complex (ABC) is added which binds to the second antibody. The peroxidase substrate TMB is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of TGFb1 present in the samples. There is cross-reactivates with TGF beta 2, TGF beta 3, TGF beta 5<1%. The purpose of this kit is the in-vitro quantitative determination of human TGFb1 in cell culture supernatants, serum, plasma (EDTA treated only) and urine samples. Pretreatment of samples with acid precipitation and neutralization step is required for optimal activity. Note: TGFbeta 1 is naturally very liable once released from its natural binding complex.
