呼吸作用是线粒体最重要的代谢。其中,线粒体中5个和呼吸相关的复合物起到了重要影响。很多呼吸链相关蛋白都是由线粒体基因组编码的。这些RNA会受到细胞核内诸如pentatricopeptide repeat (PPR) 蛋白的转录后修饰。玉米defective kernel 10(dek10) 是一个典型的籽粒变小发育迟缓的突变体。通过图位克隆发现,dek10编码了一个定位于线粒体的E族PPR蛋白。测序表明dek10突变体是由于nad3-61,nad3-62和cox2-550位点的C-U变化导致的。这些位点的变化导致Nad3显著下降,Cox2蛋白的缺失。有趣的事,复合物I并没有变化,但是NADH脱氢酶活性显著下降。复合物IV的含量显著下降。转录组测序与透射电镜(TEM)分析表明,nad3和cox2对线粒体功能密切相关。这些结果表明,E族PPR蛋白dek10对nad3和cox2的位点编辑至关重要,而这在植物发育过程中是必不可少的。 该研究中RNA-seq服务由伯豪生物提供。

烟雾病(MMD)是一种原因不明、慢性进行性的脑血管闭塞性疾病,主要表现为单侧或双侧颈内动脉远端大脑中动脉和大脑前动脉近端狭窄或闭塞伴脑底部和软脑膜烟雾状、细小血管形成。临床表现主要有脑缺血、脑出血及癫痫等。目前对于MMD的发病机理还不清楚,因此,挖掘与MMD发病相关的功能基因有助于MMD的诊断与治疗。本研究从lncRNA和mRNA出发,运用共表达分析方法,挖掘了与MMD相关的lncRNA,为理解lncRNA在MMD中的作用提供了基础。该研究中SBC human lncRNA array服务由伯豪生物提供。

皮肤粘膜淋巴结综合症(muco-cuta-meous lymph node syndrome,MCLS)又称川崎病(Kawasaki disease),是一种以全身血管炎变为主要病理的急性发热性出疹性小儿疾病。虽然已有的研究已经发现一些血来源的蛋白分子或者miRNA有潜在的诊断能力,但是,其有效性还有待提高。外泌体,一种包含了细胞特异性信息的囊泡结构,保护了内部的分子免受胞外酶类的降解。因此,通过检测的外泌体中的遗传物质,是寻找可靠诊断biomarker的理想方式。本研究从检测血清外泌体中的miRNA入手,筛选了可用于KD特异性诊断biomarker的潜在分子。该研究中miRNA表达谱芯片服务由伯豪生物提供。
