PMPA is a selective inhibitor of NAALADase with Ki value of 275 pM [1].NAALADase also known as GCPII is a zinc metalloenzyme that resides in membranes and involves in the catalyzing process of neuropeptide NAAG to NAA and glutamate. It has been shown that NAALADase has the highest expression in nervous/ prostatic tissues and cancers and NAALADase inhibition produces a variety of effects on providing neuroprotection, detection, imaging and treatment of prostate cancer [2] [3].PMPA is a potent NAALADase inhibitor and has a more activity than reported NAALADase inhibitor ZJ43. In male C57/Bl mice model, high doses of PMPA inhibited the morphine tolerance development (resembling the effect of 7.5 mg/kg of the NMDA receptor antagonist, memantine) while had no effect on severity of withdrawal; 100 mg/kg PMPA also significantly potentiated morphine withdrawal, but inhibited both acquisition and expression of morphine-induced conditioned place preference which suggested NAALADase involves in phenomena related to opioid addiction [4]. PMPA treatment increased the mice latency to enter the dark box during the training day and at the dose of 150 mg/kg PMPA treatment impaired spontaneous alternation and reduced locomotion in Y-maze task which demonstrated that PMPA affected mice learning and memory tasks through NAALADase inhibition [5].References: [1]. 第一步,拨掉ika移液器吸头弹射器放在盛放盘内;
第二步,将ika移液器容量值调至最大,拧开白套筒与手柄之间的连接螺帽;
第三步,白套筒端口向下,轻轻提起手柄放入盛放盘内,按住活塞尾部,将白套筒端口向上,置于盛放盘上部,小心取出活塞组件,以免弄掉零件;
第四步,从活塞杆上或白套筒内取出密封圈,放入盛放盘内。
最后,安装ika移液器,安装移液器的步骤与拆卸正好相反。需要注意的是,密封圈的黑色部份朝向白套筒,白色部份朝向手柄。
密封良好的移液器,只要调节机构经过校准并且可以稳定保持精度,就不必担心有问题。
如题,比如1000微升的枪,最大量程是1000,最大值要更大一些,一直用枪也没有注意过这个问题,记得原来上课老师讲是要调到最大量程,为了是弹簧处于松弛状态,那最大值应该才是最松弛状态吧,百度了一下也是有人说最大值,有人说最大量程,有人说没啥影响,想问问各位大神对这个小问题的理解,谢谢

2,计划购买100ul-1000ul移取量的移液器,目前联系了两家,一家是“瑞宁”,据说是梅特勒旗下公司,只生产移液器,比较专业;另一家是“普兰德”,据说是默克旗下产品,价格都差不多,有对该品牌了解的战友不妨谈谈注意事项?

1)按到第一档,垂直进入液面几毫米。
2)缓慢松开控制按钮,否则液体进入吸头过速会导致液体倒吸入移液器内部吸入体积减少。
3)打出液体时贴壁并有一定角度,先按到第一档,稍微停顿1s后,待剩余液体聚集后,再按到第二档将剩余液体全部压出。
二、黏稠或易挥发液体的移取
在移取黏稠或易挥发的液体时,很容易导致体积误差较大。为了提高移液准确性,建议采取以下方法:
1)移液前先用液体预湿吸头内部,即反复吸打液体几次使吸头预湿,吸液或排出液体时最好多停留几秒。尤其对于移取体积大的液体(如1000~1),建议将吸头预湿后再移取。
2)采用反相移液法:吸液时按到第二档,慢慢松开控制按钮,打液时按到第一档即可,部分液体残留在吸头内。
三、常见的错误操作
1)吸液时,移液器本身倾斜,导致移液不准确(应该垂直吸液,慢吸慢放)。
2)装配吸头时,用力过猛,导致吸头难以脱卸(无需用力过猛,选择与移液器匹配的吸头)。
3)平放带有残余液体吸头的移液器(应将移液器挂在移液器架上)。
4)用大量程的移液器移取小体积样品(应该选择合适量程范围的移液器)。
5)直接按到第二档吸液(应该按照上述标准方法操作)。
6)使用丙酮或强腐蚀性的液体清洗移液器(应该参照正确清洗方法操作)。向左转|向右转
