USAGE of the ELISA Kit DIY Materials for Hepatocyte Growth Factor (HGF)
1. Coat the plates with 100μL per well of working solution of Capture Antibody.incubate overnight at 4°C or incubate at 37°C for 2 hours.2. Aspirate and wash 1 time.3. Block the plates with 200 μL per well of working solution of Blocking Buffer. Incubate at 37°C for 1.5 hours.4. Aspirate and wash 1 time. The plates are now ready for sample detection, the protocol is the same as regular ELISA.
STORAGE of the ELISA Kit DIY Materials for Hepatocyte Growth Factor (HGF)
Antibodies, Standard and Streptavidin-HRP should be stored at -20°C. TMB should be stored at 4°C. 96-well Plate could be stored at room temperature. The contents are valid for twelve months. They are stable for one month after opening when stored at 4°C.

                                                    共价调节酶(covalent regulatory enzyme) 是一类由其它酶对其结构进行可逆共价修饰,使其处于活性和非活性的互变状态,从而调节酶活性。共价调节酶一般都存在相对无活性和有活性两种形式,两种形式之间互变的正、逆向反应由不同的酶催化。磷酸化是可逆共价修饰中最常见的类型。因为信号激酶能作用于很多靶分子,通过磷酸化作用信号能被极大地放大。蛋白激酶的调节作用能被催化水解磷酸基团的蛋白质磷酸酶逆转。通过磷酸化和脱磷酸化作用,使酶在活性形式和非活性形式之间互变。
别构调节:酶分子的非催化部位与某些化合物可逆地非共价结合后发生构象的改变,进而改变酶活性状态,称为酶的别构调节。有些酶分子在空间至少有两个不同的部位,一个为催化部位,一个为调节部位。某些物质可以与这种酶的调节部位相互作用而使酶分子构象发生改变,进而使催化部位受到影响,导致酶的催化活性改变,这种现象称为酶的别构调节,或称别位调节、变构调节
根据磷酸化的底物不同,可将蛋白激酶分为组蛋白蛋白激酶、酪蛋白蛋白激酶等,但由于蛋白激酶可磷酸化底物的多样化,这种分法很不确切,已经被根据底物磷酸化氨基酸的分类方法所取代,有些如酪蛋白蛋白激酶,只是由于习惯而一直被沿用下来。根据有无调节物将蛋白激酶分为信使依赖的蛋白激酶和非信使依赖的蛋白激酶,有些信使依赖的蛋白激酶的首字母缩略词已为人们所接受,如cAMP依赖的蛋白激酶PKA、钙和磷脂依赖的蛋白激酶PKC以及钙依赖钙调素不依赖的蛋白激酶CDPK等,它们彼此间存在结构和功能上的相关关系。
也有人认为:蛋白激酶在信号转导中主要作用有两个方面:其一是通过磷酸化调节蛋白质的活性,磷酸化和去磷酸化是绝大多数信号通路组分可逆激活的共同机制,有些蛋白质在磷酸化后具有活性,有些则在去磷酸化后具有活性;其二是通过蛋白质的逐级磷酸化,使信号逐级放大,引起细胞反应.
胶原酶按其存在的方式不同可分为人体内源性胶原酶和药用胶原酶两种。人体内源性胶原酶是指人体内部本身所具有的胶原酶,如牙龈、触膜等上皮组织和关节滑膜、椎间盘内都不同程度的存在着这种胶原酶,它在体内胶原蛋白的分解过程中发挥着不可或缺的作用。药用胶原酶是指利用生物制药的高科技手段从溶组织梭状芽孢杆菌的发酵液中提取、纯化并精制而得的白色或类白色无菌冻干粉针生物制剂。
