- Includinganoriginofreplication(i.e.theE.colioriCregion)intoacircularDNAmoleculeisamechanismtohaveanextrachromosomalelementintheprokaryoticcell.

- Inorganismswithmorethanonechromosome(eukaryotes)thereareavarietyofmechanismstoensurethatpropersegregationofchromosomesoccurs,i.e.tomakesurethatdaughtercellscontainequalnumbersofallthechromosomes.
- Onebasicmechanismisthateachchromosomecontainsessentialgenes,andifthesearelost,thecellcannotsurvive.
Drugresistance- Byfarthemostcommonapproachtothemaintenanceofplasmidsisthroughtheincorporationofdrugresistancegenes.
- Thesearealsoknownasselectablemarkers,i.e.wecanselectfortheirpresencebyincludingantibioticsinthegrowthmedia.
Ampicillin- Ampicillinbindstoandinhibitsanumberofenzymesinthebacterialmembranethatareinvolvedinthesynthesisofthegramnegativecellwall.
- Therefore,propercellreplicationcannotoccurinthepresenceofampicillin.

Tetracycline
- Tetracyclinebindstoaproteinofthe30SsubunitoftheribosomeandinhibitsribosomaltranslocationalongthemessengerRNAwhichcodesfortheprotein(i.e.thedruginterfereswithnormaltranslationorproductionofproteins).
- Thetetracyclineresistancegene(tetr)encodesa399aminoacidoutermembraneassociatedproteinofgramnegativecellsthatpreventstheantibioticfromenteringthecell.
- Thus,thisdrugresistancegenedoesnotdestroytheantibiotic.Pressurewillbemaintainedthroughoutthecellcultureprocesstokeeptheplasmidcontainingthedrugresistantgene.
Chloramphenicol- Chloramphenicolbindstotheribosomal50Ssubunitandinhibitsproteinsynthesis.
- Thechloramphenicolresistancegene(Cmr)codesforaproteinknownatthecatprotein.
- Thecatproteinisatetramericcytosolicproteinthat,inthepresenceofacetylcoenzymeA,catalyzestheformationofhydroxylacetoxyderivativesofchloramphenicolthatareunabletobindtotheribosome.
- Likewithampicillin,theCmrgeneproductdestroystheantibiotic.
- Additionally,theexpressionofcatproteinisinfluenced(downregulated)bythepresenceofglucoseinthemedia.
Kanamycinandneomycin- Bindtoribosomalcomponentsandinhibitsproteinsynthesis.

- IftheColE1oriregiondoesnothavetheropgene,thentheresultingplasmidismaintainedwithanaveragecopynumberof100-200plasmids/cell.Thisisconsideredahighcopynumbertypeofplasmid.
