Detection of Human HSP90 alphaby Western Blot.Western blot shows lysates of HEK293 human embryonic kidney cell line. PVDF membrane was probed with 0.5 µg/mL of Rabbit Anti-Human HSP90a Antigen Affinity-purified Polyclonal Antibody (Catalog # A-440) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for HSP90a at approximately 85-95 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: HSP90 alpha
The heat shock protein-90 kDa (HSP90) is a composite name for a large group of genes whose molecular weights average 90 kDa. HSP90 functions primarily as a molecular chaperone, facilitating the folding of other cellular proteins, preventing protein aggregation, or targeting improperly folded proteins to specific degradative pathways. In addition to its role as a molecular chaperone and stress response protein, HSP90 is a central component in a number of cellular processes including hormone signaling and cell cycle control. HSP90 is ubiquitously expressed, highly conserved, and accounts for 1-2% of the total cellular protein. Eukaryotic cells have two principal forms of HSP90 including the inducible HSP90 alpha(also known as HSP90AA1, HSP90A, HSPC1, HSPCA and HSP86) and the constitutively expressed HSP90 beta(also known as HSP90AB1, HSP90B, HSPCB, HSPC2, and HSP89-beta. While HSP90 alphaand HSP90 betashare 90% identity, this affinity-purified polyclonal antibody is specific for HSP90 alpha.

过完年,再开培养箱参数不稳定,两个箱子,co2数值一直在闪~我要养细胞,参数设置的是37℃,5%CO2,请问各位大神有知道咋回事的~还有面板上各种符号啥意思?图中标记的都是在闪的参数
对细胞进行一定的药物处理后,还要在37度中孵育培养24到48小时不等,还能放入二氧化碳培养箱中进行培养吗?
一般电热恒温培养箱是:RT+5~65℃
生化培养箱带有制冷装置,一般是:0~60℃,无需高于环境温度
2. 我的经验是在补充水时最好把水加热一下,达到培养箱的温度。
3. 这么早就养细胞了,我们这里一般是用70%的乙醇擦洗,然后再用0.1%的新洁尔灭擦洗,不放心的话再用高锰酸钾加甲醛1:1比例熏一遍。注意要是熏的话千万注意混合之后马上离开,然后密闭细胞培养室,味道很刺激的。最好在养细胞前消毒,否则,确实没地方放细胞。
4. 我们的方法比较简单,先用75%酒精擦拭内壁,通风10分钟,紫外线照射30分钟即可。
5. 我们的通常处理是先用75%的酒精擦拭内壁,然后用甲醛+高锰酸钾熏蒸一个晚上,然后通风一整天(同时打开紫外线照射)。
6. 我们是把培养箱里的板层拿出来高压一下,箱壁用75%酒精擦一遍,换用诗乐氏擦一遍,再紫外照一夜,效果很好,其他的请高手指教。
7. 熏蒸后吹一天是绝对不够的,最起码要一个星期才可以让甲醛散尽,要不细胞长不好的
8. 有的培养箱能加热内壁到90度,也有的带有紫外线功能。常规应该每周用酒精擦洗内部一次。具体的操作请参考培养箱的手册,或着咨询培养箱技术支持。使用消毒剂应小心,培养箱内有检测温度、湿度和二氧化碳浓度的传感器,腐蚀性消毒剂或有机溶剂有可能损坏某些器件,请慎用。另外,挥发性的甲醛有毒,易燃、易爆。如果在培养箱内残留甲醛的话,细胞会死光光哦(我实验室的真实故事)。总之,先看培养箱手册,再联系技术支持,方可万无一失啊。
9. 常规最好什么也不放!硫酸铜的作用是抑制细菌生长(但多数用在处理污染的孔板)。因为现在的孵箱都有抑菌功能。
10. 我们实验室的做法:把平皿高压灭菌后,在无菌操作台内加入适量的灭菌双蒸水或者生理盐水.再加少量新洁尔灭,浓度没关系.细胞养的都可以.
