Q1: What is the lower limit and minimal amount of DNA that can be recovered?
Picogram levels of DNA can be recovered. The limitation is based on sensitivity of detection method.
Q2: How to process naked DNA stored in DNA/RNA Shield?
Use the standard protocol (add 2 or 5 volumes of DNA binding buffer depending on DNA size).
Q3: What to do if ethanol addition to the DNA Wash Buffer was omitted?
The DNA will be eluted off the column. Rebind samples using the appropriate amount of DNA Binding Buffer and wash the column with the properly prepared wash buffer.
Q4: What happens if more DNA was loaded on the columns than the stated maximum binding capacity?
Oversaturation of the column can result in total DNA loss due to clogging of silica matrix.
Q5: How many times can columns be reloaded?
We recommend no more than 5 times as binding efficiency might decrease.
Q6: What is the minimum input volume of DNA sample?
Working with volumes below 50 µl can result in decreased recovery. We recommend raising the starting volume to 100 µl with water to ensure optimal binding conditions.

刚对实验室的通风柜做了个气流流向测试,报告出来了不知道该怎么管理这个文件了,当做设备确认方案?感觉不是很合理。放公司的安环部门?他们也没有档案室。我该怎么保存这个文件呢?
一连串的涨工资,补助等等跟钱相关的郁闷事就不好在这里说了,总之我估计,虽然都比以前多了,但是真正没几个心里舒服的,原因,就问那些有奇怪想法的决策人吧.表面上看,我应该开心,但实际也没觉得应该开心到哪去.
提及本人那"恐怖"的工作,想必看过有关"通风橱"日志的朋友们应该相对了解的多一些,恩,药物合成.电视上都看过实验室瓶瓶罐罐的吧,本人工作环境虽赶不上那么高端,但实质都是这些东西.最近呢,为之困扰的是这次项目中的一个试剂,属于剧**类试剂,含巯基化合物,搞化学的人都知道,巯基可是不得了的,毒性咋先不说,单是这味道,是很不好闻的,臭,都臭不明白!用某些人的话说,臭,都不是个正宗的臭味,结果上网一查,该化合物的嗅觉最低阈值是相当的低,解释起来就是说,极少的极微量的液体就会引起很大的气味.
味,咱做合成的人,倒不怕.用过那么多试剂了,比这吓人的,"化肉化骨水"之类的都用着跟玩似的了,也没什么.可是这东西不同寻常啊,它太臭了做一次实验,如果真的不带防护措施的话,我一天的饭都不想吃,想起了就想吐,所以必须,跟领导提出买全套防毒设施.包括防毒面具和防护服.
领导还算重视,特批,买!
结果呢?跟采购提了计划,本以为这东西,如果上个心,不难买的,谁知道呢,采购也是隔行如隔山,怎么说也不懂,怎么给找网上的图片也不行.也是我自己用,没人着急,今天到早上扫完雪我就跑去做实验,一直站到下午3点,腿都软了,跑回办公室还得查这些,不单是查销售厂家,还得打电话询问,可是除我之外没人着急,可是除我之外没人闻味了
这些呢,也不算什么,累就累点儿,也都是给自己干活儿,谁让咱是个认真的人!结果呢!上网一查才知道,如果达到我所形容的防护级别,也就是全身防护,且尽量不闻到任何气味,就需要那种背氧气瓶的全套密闭装备,钱贵不说,得至少1万,就这套设备穿身上也够我戗了,工作不便捷,我自己也不想.无奈,只能退而求其次,结果各种选择都不太能两全其美,本来这边买防护设施着急,我那边实验还得有人做,做的不顺利也着急,那会儿真想放弃,我自己都打心眼里不想买什么防护设施了,不就是毒么?我这么累这么忙,我还得象求着谁了似的,我图个什么啊!就觉得太委屈了!加之前一阵子各种跟钱有关的风波,那就不能想了,再想只是给自己添烦恼了.
其实困难最主要在于,我从来没接触过这些防毒设施,对于各个样式的防护服和防毒面具,不知道其防护特点是否正是我所需要的.网上图片毕竟是太平面化,跟实际物品的差距很大.而身边也没有参考和推荐,时间有限,感觉,很懊恼.
实际上,可以自己做项目的感觉很好,起码是给自己一个锻炼的机会,当然,也是对自己负责任,可是一连串的事件实在是给我冲击很大,心理的困难很重.这样的情况下,人会觉得太过于失衡.自己知道,同样的劳动报酬下,工作量不同,不是在我这个年龄应该去比的,或者你多做的,也是多得的.但是别往我这工作的激情与热情上面泼冷水吧!,,,无奈,,,
允许我暂时的牢骚,可能明天的我,又会开始象没受过"伤害"一样辛勤的奋战在我的"有毒战场",也希望大家不要一闻着臭的东西就来追问是不是我又开始做实验,最好别因此而隔离我,阿门.
