ErbB4,alsocalledHer4(humanepidermalgrowthfactorreceptor4),isatypeImembraneglycoproteinthatisamemberoftheErbBfamilyoftyrosinekinasereceptors.ErbBfamilymembersserveasreceptorsfortheepidermalgrowthfactor(EGF)familyofgrowthfactors.ErbB4isexpressedinnormalskeletalmuscle,heart,pituitary,brainandseveralbreastcarcinomas.ErbB4ligandsincludetheneuregulins,beta-cellulinandheparin-bindingEGF-likegrowthfactor(HB-EGF).MonomericErbB4bindsitsligandswithlowaffinity.Typically,heterodimerizationwithErbB2formsthehighaffinityreceptorcomplex.However,ErbB4hasalsobeenshowntoheterodimerizewithbothErbB1andErbB3.Ithasbeensuggestedthattheidentityoftheligandmayinfluencethedimerizationpartner.SeveralErbB4isoformsexist.TwoofthesedifferinthepresenceofjuxtamembraneextracellularsequenceswhichregulatetheabilityofTACE(TNF-alphaconvertingenzyme,ADAM17)toproteolyticallycleaveErbB4fromthecellsurface.Theseisoformsexhibittissue-specificexpression.Anotherisoformlacksthephosphoinositide3-kinaseactivationsequencepresentintheErbB4cytoplasmicdomain.HumanErbB4consistsof1308aminoacids(aa)witha25aasignalsequence,a626aaextracellulardomain,a24aatransmembraneregion,anda633aacytoplasmicdomain.ErbB4appearstoplayimportantrolesinneuronaldevelopment,developmentoftheheartandcancer.
