Abstract
Tolerogenicdendriticcells(DCs)caninduceregulatoryTcellsanddampenpathogenicTcellresponses.Therefore,theyare
possibletherapeutictargetsinautoimmunediseases.InthisstudyweinvestigatedwhethermousetolerogenicDCsare
inducedbythephytonutrientcarvacrol,amoleculewithknownanti-inflammatoryproperties,binationwitha
physiologicalstress.WeshowthattreatmentofDCswithcarvacrolandthermalstressledtothemRNAexpressionofboth
pro-andanti-inflammatorymediators.Interestingly,treatedDCswiththismixedgeneexpressionprofilehadareduced
abilitytoactivatepro-inflammatoryTcells.Furthermore,theseDCsincreasedtheproportionofFoxP3+regulatoryTcells.In
vivo,prophylacticinjectionofcarvacrol-thermalstresstreatedDCspulsedwiththediseaseinducingantigenwasableto
suppressdiseaseinamousemodelofarthritis.ThesefindingssuggestthattreatmentofmousebonemarrowderivedDCs
withcarvacrolandthermalstressinduceafunctionallytolerogenicDCthatcansuppressautoimmunearthritis.Herewith
carvacrolseemstooffernovelopportunitiesforthedevelopmentofadietarybasedinterventioninchronicinflammatory
diseases.
