Epidermalgrowthfactor(EGF)isthefoundingmemberoftheEGFfamilythatalsoincludesTGF-alpha,amphiregulin(AR),betacellulin(BTC),epiregulin(EPR),heparin‑bindingEGF‑likegrowthfactor(HB‑EGF),epigen,andtheneuregulins(NRG)-1through-6(1).MembersoftheEGFfamilyshareastructuralmotif,theEGF‑likedomain,whichischaracterizedbythreeintramoleculardisulfidebondsthatareformedbysixsimilarlyspacedconservedcysteineresidues(2).AllEGFfamilymembersaresynthesizedastypeItransmembraneprecursorproteinsthatmaycontainseveralEGFdomainsintheextracellularregion.Thematureproteinsarereleasedfromthecellsurfacebyregulatedproteolysis(1).The1207aminoacid(aa)humanEGFprecursorcontainsnineEGFdomainsandnineLDLRclassBrepeats.Thematureproteinconsistsof53aaandisgeneratedbyproteolyticexcisionoftheEGFdomainproximaltothetransmembraneregion(3).MaturehumanEGFshares70%aasequenceidentitywithmaturemouseandratEGF.EGFispresentinvariousbodyfluids,includingblood,milk,urine,saliva,seminalfluid,pancreaticjuice,cerebrospinalfluid,andamnioticfluid(4).FourErbB(HER)familyreceptortyrosinekinasesincludingEGFR/ErbB1,ErbB2,ErbB3andErbB4,mediateresponsestoEGFfamilymembers(5).Thesereceptorsundergoacomplexpatternofligandinducedhomo-orhetero-dimerizationtotransduceEGFfamilysignals(6,7).EGFbindsErbB1anddependingonthecontext,inducestheformationofhomodimersorheterodimerscontainingErbB2.Dimerizationresultsinautophosphorylationofthereceptoratspecifictyrosineresiduestocreatedockingsitesforavarietyofsignalingmolecules(5,8).BIOLOGicalactivitiesascribedtoEGFincludeepithelialdevelopment,angiogenesis,inhibitionofgastricacidsecretion,fibroblastproliferation,andcolonyformationofepidermalcellsinculture.

EGFR was detected in immersion fixed frozen sections of&amp;nbsp;mouse embryo (13&amp;nbsp;d.p.c.) using Goat Anti-Mouse EGFR Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1280) at 15&amp;nbsp;µg/mL overnight at 4&amp;nbsp;°C. Tissue was stained using the Anti-Goat HRP-DAB Cell &amp;amp; Tissue Staining Kit (brown; Catalog&amp;nbsp;# CTS008) and counterstained with hematoxylin (blue). 抗体都是免疫球蛋白而免疫球蛋白不一定都是抗体。原因是抗体是由浆细胞产生,且能与相应抗原特异性结合发挥免疫功能的球蛋白。而免疫球蛋白是具有抗体活性或化学结构与抗体相似的球蛋白,如骨髓瘤患者血清中异常增高的骨髓瘤蛋白,是由浆细胞瘤产生,其结构与抗体相似,但无免疫功能。因此,免疫球蛋白可看做是化学结构上的概念,抗体则是生物学功能上的概念。
