Description:
RedBloodCell(RBC)LysisBufferhasbeendesigned,formulated,andtestedtoensureoptimallysisofRBCsinsingle
cellsuspensionswithminimaleffectsonleukocytes.RBCLysisBufferissuppliedasa10Xsolutioncontaining
ammoniumchloride,potassiumcarbonate,andEDTA,andshouldbedilutedindeionizedwaterpriortouse.This
buffercanbeusedforlysisofhuman,mouse,rat,rabbitandnon-humanprimatesamples.

Preparation:
Thisredbloodcell(RBC)lysisbufferissuppliedasa10Xsolutionandshouldbedilutedto1Xindeionizedwater.The
pHofthe1XsolutionshouldfallwithintherangeofpH7.1-7.4.AdjustthepHifnecessary.Warmthe1Xsolutionto
roomtemperaturepriortouse.

LysisofHumanPeripheralBloodRBCs:
1.Dilutethe10XRBCLysisBufferto1Xworkingconcentrationwithdeionizedwater.Warmthe1Xsolutionto
roomtemperaturepriortouse.
2.Add2.0mlof1XRBCLysisBuffertoeachtubecontainingupto100µlofwholeblood.
3.Gentlyvortexeachtubeimmediatelyafteraddingthelysingsolution.Incubateatroomtemperature,protected
fromlight,for10-15minutes.
4.Centrifuge1000rpmfor5minutes.Aspiratesupernatantwithoutdisturbingpellet.
5.Resuspendthepelletintheappropriatebuffer(e.g.,4abioCellStainingBuffer,Cat.No.FXP005),wash1X.
6.Resuspendandproceedwithfurtherprocedures.
