Tissue-typeplasminogenactivator(tPA)isa68-kDaserineproteasethatconvertsthezymogenplasminogenintotheactiveserineproteaseplasmin,whichdigestsfibrinandinducesthedissolutionoffibrinclots(1).tPAissynthesizedbyendothelialcellsinnormalbloodvesselsanddisplaysrelativelyhighaffinityforfibrin,suggestingthatitfunctionspredominatelyinphysiologicalthrombolysisinvivo(2).AhighleveloftPAisagoodprognosticMarkerforbreastcancer(3).tPAmayminimizetheformationofmetastasisbypreventingtumorcelladherenceatsitesoftrauma(4).Ontheotherhand,gastrointestinalcancerisaccompaniedbyadecreaseintPA(5).

TheAssaySenseHumantPAChromogenicActivityKitisdevelopedtodeterminehumantPAactivityinplasmaandcellculturesupernatants.TheassaymeasurestheabilityoftPAtoactivatetheplasminogentoplasminincoupledorindirectassaysthatcontaintPA,plasminogen,andaplasmin-specificsyntheticsubstrate.Theamountofplasminproducedisquantitatedusingahighlyspecificplasminsubstratereleasingayellowpara-nitroaniline(pNA)chromophore.ThechangeinabsorbanceofthepNAinthereactionsolutionat405nmisdirectlyproportionaltothetPAenzymaticactivity.
