IntendedUse:TheInfluenzaVirusBIgG-ELISAisintendedforthequalitativedeterminationofIgGclassantibodiestoInfluenzaVirusBinhumanserumorplasma(citrate).
Introduction:InfluenzaareRNAvirusesofthefamilyOrthomyxoviridae.Influenzavirusesaredividedintothreetypes,designatedA,B,andCwhicharedifferentiatedbythespecificityofasolubleantigenassociatedwiththeinternalribonucleoproteincomponentofthevirion.Thevirionsarespericalparticlesof80-120nmindiameterconsistingoftheribonucleoproteincomponentandenvelopedbymatrix
proteinandalipidbilayerwhichcontainstwospikelinestructures:viralhemagglutinin(H)andviralneuraminidase(N).
Influenzavirusesarerespiratorytractpathogenswhicharetransmittedbydirectcontact,large-dropletinfection,orbycontaminatedsurfaces.InfluenzatypesAandBareresponsIBLeforepidemicsofrespiratoryillnessthatoccuralmosteverywinterandareoftenassociatedwithincreasedratesofhospitalizationanddeath.TypeCinfectionsusuallycauseeitheraverymildrespiratoryillnessornosymptomsatall;itdoesnotcauseepidemicsanddoesnothavetheseverepublichealthimpactthatinfluenzatypesAandBdo.
PandemicsofinfluenzavirustypeAinfectionshaveoccuredat10to20yearintervalssince1890.ApparentlythisresultsfromalterationsinthecompositionoftheHandNantigens(antigenicdriftandantigenicshift).CurrentlytwosubtypesofinfluenzaAarecirculatingworldwide.Normallyinfluenzaisaself-limitingdiseaselastingfor3to7days,butsomepeopledevelopseriousandpotentiallylife-threateningmedicalcomplications,suchaspneumonia,particularlyinchildren,elderlypeopleandothervulnerablegroups.

Principlesoftheassay:ThequalitativeimmunoenzymaticdeterminationofIgG-classantibodiestoInfluenzaVirusBisbasedontheELISA(Enzyme-linkedImmunosorbentAssay)technique.
MicrotiterstripwellsareprecoatedwithInfluenzaVirusBantigenstobindcorrespondingantibodiesofthespecimen.AfterwashingthewellstoremoveallunboundsamplematerialhorserADIshperoxidase(HRP)labelledanti-humanIgGconjugateisadded.ThisconjugatebindstothecapturedInfluenzaVirusBspecificantibodies.TheimmunecomplexformedbytheboundconjugateisvisualizedbyaddingTetramethylbenzidine(TMB)substratewhichgivesabluereactionproduct.TheintensityofthisproductisproportionaltotheamountofInfluenzaVirusBspecificIgGantibodiesinthespecimen.Sulphuricacidisaddedtostopthereaction.Thisproducesayellowendpointcolour.Absorbanceat450nmisreadusinganELISAmicrowellplatereader.
StorageandStABIlity:Thereagentsarestableuptotheexpirydatestatedonthelabelwhenstoredat2...8°C.
LimitationsoftheTest:Bacterialcontaminationorrepeatedfreeze-thawcyclesofthespecimenmayaffecttheabsorbancevalues.Diagnosisofaninfectiousdiseaseshouldnotbeestablishedonthebasisofasingletestresult.Aprecisediagnosisshouldtakeintoconsiderationclinicalhistory,symptomatologyaswellasserologicaldata.
Inimmunocompromisedpatientsandnewbornsserologicaldataonlyhaverestrictedvalue.
