ProteinCisavitaminK-dependentplasmaantithromboticandanti-inflammatoryzymogenicglycoproteinthatissynthesizedintheliver.ProteinChasalightchainof155aminoacids(21kDa)andaheavychainof262aminoacids(41kDa)linkedbyadisulfidebond.Onendothelialcellmembrane,thrombin-thrombomodulincomplexcleavesa12-residuepeptidefromproteinCaminoterminusoftheheavychainandconvertsittoactivatedproteinC(APC).APCinactivatescoagulationFactorVaandFactorVIIIaandperformsamajorroleinregulatingbloodclotting,inflammation,andapoptosis(1-3).ProteinCdeficiencycausesneonatalpurpurafulminans,thrombophilia,andrecurrentvenousthrombosis(4-6).ProteinCpathwaycomponentshavebeenstudiedinthetreatmentofcomplexdisorders,includingseveresepsis,thrombosis,andischemicstroke(7).
PrincipleoftheAssay
TheAssayMaxHumanProteinCELISA(Enzyme-LinkedImmunosorbentAssay)kitisdesignedfordetectionofhumanProteinCinplasmaandserum.ThisassayemploysaquantitativecompetitiveenzymeimmunoassaytechniquethatmeasureshumanproteinCinlessthan3hours.ApolyclonalantibodyspecificforhumanproteinChasbeenpre-coatedontoa96-wellmicroplatewithremovablestrips.ProteinCinstandardsandsamplesiscompetedwithabiotinylatedproteinCsandwichedbytheimmobilizedantibodyandstreptavidin-peroxidaseconjugate.Allunboundmaterialisthenwashedawayandaperoxidaseenzymesubstrateisadded.Thecolordevelopmentisstoppedandtheintensityofthecolorismeasured.
