
Overview:
MEKK6 or MAP3K6 (mitogen-activated protein kinase kinase kinase 6) is a member of the serine/threonine protein kinase family. MEKK6 was identified by its interaction with MAP3K5/ASK, a protein kinase and an activator of c-Jun kinase (MAPK7/JNK) and MAPK14/p38 kinase (1). MEKK6 cascades relay signals from the cell surface to the nucleus which result in gene transcription (2). MEKK6 activates the JNK and p38 pathway and induces apoptosis when overexpressed in stably transfected cells. MEKK6 transcript is expressed in several tissues, with the highest expression in heart and skeletal muscle.

也有人认为:蛋白激酶在信号转导中主要作用有两个方面:其一是通过磷酸化调节蛋白质的活性,磷酸化和去磷酸化是绝大多数信号通路组分可逆激活的共同机制,有些蛋白质在磷酸化后具有活性,有些则在去磷酸化后具有活性;其二是通过蛋白质的逐级磷酸化,使信号逐级放大,引起细胞反应.
