The combined solution is ideal for protein gel applications.1X Red Loading Buffer Composition: 187.5 mM Tris-HCl (pH 7.0), 6% SDS (w/v), 30% glycerol, 0.03% phenol red (w/v)30X Reducing Agent: 1.25 M DTTSDS interacts with positively charged amino acids of proteins, thereby disrupting interactions that form protein structures to separate proteins based on size rather than charge or shapeDTT is present to help reduce structural disulfide bonds Prepare fresh 3X Reducing Red Loading Buffer by adding 1/10 volume 30X Reducing Agent to 1 volume of 3X Red Loading Buffer. Prepare samples by adding 2 volumes of Sample to 1 volume 3X Reducing Red Loading Buffer (from step 1). Heat samples to 95-100 C for 3-5 minutes. After a quick microcentrifuge spin, load directly on to a gel. To ensure uniform mobiity, load an equal volume of 1X Reducing Blue Loading Buffer into any unused wells. Reagents stored at different temperatures. DTT will degrade at room temperature. Keep the two components separate at store at their proper temps. Quality Control tests are performed on each new lot of NEB product to meet the specifications designated for it. Specifications and individual lot data from the tests that are performed for this particular product can be found and downloaded on the Product Specification Sheet, Certificate of Analysis, data card or product manual.
