连接酶有T4噬菌体DNA连接酶、T4噬菌体RNA连接酶、大肠埃希菌DNA连接酶等。DNA连接酶可连接平端,也连接粘端。反应需有Mg2+和ATP存在,pH7.5-7.6。最适温度37℃,30℃以下活性明显下降,但考虑到被连接DNa 的稳定性和粘性末端的退火温度,一般平端连接用20-25℃,粘端连接用12℃左右。
聚合酶有DNA聚合酶(以DNA为模板合成DNA大肠埃希菌DNA聚合酶Ⅰ,大肠埃希菌DNA聚合酶Ⅰ大片段(Klenow大片段),T4或T7噬菌体DNA聚合酶等);RNA聚合酶(以DNA为模板合成RNA,T7或T3噬菌体RNA聚合酶);逆转录酶(以RNA为模板合成DNA,除RNA病毒中发现外,发现大肠埃希菌DNA聚合酶Ⅰ和Taq DNA聚合酶都有逆转录活性)。

在哺乳动物组织中,节律基因的表达受到局部的振荡器或视交叉上核中生物钟主钟控制的系统信号的调控。本研究发现是受刺激的体温循环而不是外周振荡器,控制纤维母细胞中的冷诱导RNA结合蛋白(CIRP)的节律性表达。反过来,功能缺失性实验表明CIRP是高振幅的节奏基因表达所需的。利用基于生物素-链亲和素的交联和免疫沉淀反应(CLIP),本研究对CIRP结合的RNA进行了全转录组分析,发现了一些编码昼夜节律振荡器蛋白的转录物,包括CLOCK蛋白。此外,在CIRP缺失的纤维母细胞中,CLOCK的含量大大减少。因为在这些细胞中,CLOCK的异位表达提高了节律基因的表达水平,因此我们猜测CIRP通过调节CLOCK的表达水平赋予了昼夜节奏振荡器的健壮性。
大家有用过Invitrogen的T4连接酶吗?说明书上是23-26度连接,一般的连接酶不都是16度吗?应该用多少度呢?另外,说明书上还说连接后为了达到更好的转化效率,应将连接反应液至少稀释5倍再转化,是这样吗?谢谢大家帮忙啊
RNA聚合酶Ⅰ存在于核仁中,转录rRNA顺序。RNA聚合酶Ⅱ存在于核质中,转录大多数基因,需要“TATA”框。RNA聚合酶Ⅲ存在于核质中,转录很少 RNA聚合酶几种基因如tRNA基因如5SrRNA基因。有些重复顺序如Alu顺序可能也由这种酶转录。
