最近做的三次WB，用的试剂都一样，条件都一样，转膜是300mA40min，分子量是40kd和45kd。三次结果如下：第一次，转膜后Marker正常，最后显影条带正常。第二次，转膜后marker不见了，最后显影条带如图，一张膜呈现一头大一头小的条带，另一张膜结果正常。第三次，转膜后还是没有marker，显影后条带上什么都没有求帮忙分析同样的条件同一瓶母液，为什么会这个样子呢……心里急得很……

想看下人肿瘤组织内巨噬细胞的浸润情况，我以为一个CD11b就够了。咨询了一下抗体公司，竟然让我订三个抗体，分别是CD11b，CD68和CD163，后两者不是M1和M2的分型吗？
另外查了下CD11b也确实在别的一些免疫细胞有所表达，请教下，有专门的特异性抗体吗？没课题没经费，只能省省省了

如图所示，用的Invitrogen10%预制胶，Marker用的生工5-245，marker一直跑不开，75以上的虽然跑开了可是已经弥散到看不清楚了，原来自己配的胶从来没有这种情况，换高级货咋还这样了？？各位高手帮助解答。会不会是电压问题？预制胶说明书上说用200V，原来BioRad跑一直用120V。总之很疑惑，期待高手解答。

连续三次都这样，Marker的泳道的位置有很多亮条带，但Marker本来的条带是不亮的。我做了内参和目的，一张胶上左右两部分分别都加样品和Marker，切开转膜分别孵育，一块儿显影，内参的Marker就不会亮，只有目的那一张Marker会亮，昨天做的借了别人的Marker，显影出来还是有那个条带。
我的目的条带还没有Marker那部分的条带亮，Marker的部分非常亮。
