Intended Use:
AESKUBLOTS Food Intolerance ONE is a membrane based enzyme immunoassay for quantitative detection of food-specific IgG4antibodies in human serum or plasma. Antigens are located as parallel lines at exactly defined positions on a nitrocellulose membrane.The assay permits a large number of tests to be conducted in parallel over a short period of time. The assay is used to support the diagnostic process in patients suspected to have food intolerances.

配制分三步: 1) 1 M Tris-HCl (pH 8.0) 50 ml的配制:称取Tris碱6.06 g,加超纯水40 ml溶解,滴加浓HCl约2.1 ml调pH至8.0,定容至50 ml。
低离子强度时,迁移率快。但离子强度过低,缓冲液的缓冲容量小,不易维持pH恒定。高离子强度时迁移率慢,因此为了获得更快的反应速度,在缓冲容量允许的范围内,离子强度应该尽可能小。
蛋白质上样缓冲液中的SDS和DTT分别可以屏蔽蛋白质电荷以及破坏二硫键,避免形成多聚体。
DNA上样缓冲液主要是起沉降和指示作用,不可用于蛋白,否则会影响蛋白相对分子量定量。
同样蛋白上样缓冲液中的tris-HCl浓度过高也会使溴酚蓝条带扭曲。
加样缓冲液的主要作用是使PCR产物与其混合,
使DNA沉于加样孔的底部,防止DNA跑出来.
由弱酸及其盐、弱碱及其盐组成的混合溶液,能在一定程度上抵消、减轻外加强酸或强碱对溶液酸碱度的影响,从而保持溶液的pH值相对稳定。这种溶液称为缓冲溶液。
