Thecadherinsuperfamilyisalargefamilyofmembrane-associatedglycoproteinsthatengageinhomotypic,calcium-dependent,cell-to-celladhesionevents.Thesuperfamilycanbedividedintoatleastfoursubfamiliesbasedonitsmember’sextracellular(EC)regionsandcytoplasmicdomains(1,2).Theseincludeclassical cadherins,desmosomalcadherins,protocadherins,andcadherin-likemoleculesthatcontainavariablenumberofECandtransmembrane(TM)domains(1).Cadherin‑6,alsoknownasKCADorK-cadherin,isaclassical cadherinof110-120kDthathasatleastonefulllengthandtwoalternatespliceformsranginginsizefrom105‑120kDa(3).Humancadherin-6issynthesizedasa790aminoacid(aa)typeItransmembraneglycoproteinthatcontainsa18aasignalpeptide,a35aaprosequence,a562aaextracellularregion,a21aatransmembranesegment,anda154aacytoplasmicdomain(4,5).TherearefiveECcadherindomainsthatareapproximately110aainlength.Thispatternisconsistentwithclassicalcadherinfamilymoleculesthataremodularintheirextracellularregionandmediatecalcium-dependentcell-to-celladhesionthroughtheirCa++-bindingrepeats(2).DuetotheabsenceofaHis-Ala-ValmotifinitsmostN-terminalcadherinrepeat,Cadherin-6canbefurtherclassifiedasatypeIIclassicalcadherin(4).Onecadherin-6splicevariant(termed6/2)showsa9aasubstitutionforthe94aathatspanresidues283to376ofthefull-lengthextracellularregion(3).Asecondsplicevariantshowsa36aasubstitutionfortheC-terminal163aaofthetransmembraneandcytoplasmicregion(6).Theextracellularregionofhumancadherin-6is98%aaidenticaltoratcadherin-6extracellularregion,plus60%and58%aaidenticaltotheextracellularregionsofhumancadherin8and11,respectively.Cadherin-6hashighexpressioninkidney,brain,andcerebellum,andlowexpressioninlung,pancreas,gastricmucosa,andcytotrophoblasts(4,5,7-9).Cadherin-6isalsofoundinrenal,lung,andovariancarcinoma(7,10).Asaclassiccadherin,cadherin-6willformhomodimersandpromoteintercellularadhesionwithitselfandpossIBLy,cadherin-9and-14(4,11).
