Notch-1isa300kDatypeItransmembraneglycoproteinthatisoneoffourNotchhomologuesinvolvedindevelopmentalprocesses(1‑3).Notchsignalingisimportantformaintainingstemcellsandinducingdifferentiation,especiallyinthenervoussystemandlymphoidtissues(2‑4).Notchcanspecifybinarycellfates.Forexample,itpromotesT-celloverB-celldevelopmentfromacommonprecursor(2).MouseNotch-1issynthesizedasa2531aminoacid(aa)precursorthatcontainsan18 aasignalsequence,a1707aaextracellulardomain(ECD)with36EGF-likerepeatsandthreeLin-12/notchrepeats(LNR),a21aatransmembrane(TM)segmentanda785aacytoplasmicdomainthatcontainssixankyrinrepeats,aglutamine-richdomainandaPESTsequence.The11thand12thEGF-likerepeats,thatbindligandssuchasJaggedandDelta-likefamiliesinhumans,correspondtoaa412‑488inmouseNotch-1(6).ElongationofO-linkedfucosechainsbyFringefamilymembersatasitewithinthisregioncaninhibittheinteractionofNotchwithJaggedligands,therebypromotingDelta-likeligandinteractions(7).TheNotch-1receptorundergoespost-translationalfurin-typeproteolyticcleavage,generatingaheterodimerthroughtheinteractionofahydrophobicareaC-terminaltotheLNRontheextracellularregionwiththetransmembrane/cytoplasmicportion(8,9).Uponligandbinding,additionalsequentialproteolysisbyTNF-convertingenzyme(ADAM17)andthepresenilin-dependentgamma-secretaseresultsinthereleaseoftheNotchintracellulardomain(NICD)whichtranslocatesintothenucleus,activatingtranscriptionofNotch-responsivegenes(10).MouseNotch-1ECDaa19‑526,whichincludesthefirst13EGFrepeats,shows94%,91%,86%and79%aaidentitywithcorrespondingregionsofrat,human,canine,andchickenNotch-1,respectively.Thisregionalsoexhibits55‑58%aaidentitywithhumanNotch-2andNotch-3.
