Ephrin-A4,alsoknownasLERK-4andEFL-4,(1)isamemberoftheephrinligandfamilywhichbindsmembersoftheEphreceptorfamily.Allligandsshareaconservedextracellularsequence,whichmostlikelycorrespondstothereceptorbindingdomain.Thisconservedsequenceconsistsofapproximately125aminoacidsandincludesfourinvariantcysteines.TheA-classligandshaveaGPIanchorfollowingtheconservedsequence.Ephrin-A4hasbeenshowntobindEphA2,EphA3,EphA4,EphA5,EphA6,EphA7,andEphB1(2,3).TheextracellulardomainsofhumanandmouseEphrin-A4share80%aminoacididentity.Onlymembrane-boundorFc-clusteredligandsarecapableofactivatingthereceptorinvitro.Whilesolublemonomericligandsbindthereceptor,theydonotinducereceptorautophosphorylationandactivation(2).Invivo,theligandsandreceptorsdisplayreciprocalexpression(3).Ithasbeenfoundthatnearlyallreceptorsandligandsareexpressedindevelopingandadultneuraltissue(3).TheEph/ephrinfamiliesalsoappeartoplayaroleinangiogenesis(3).
