HGFR,alsoknownasMet(fromN-methyl-N’-nitro-N-nitrosoguanidineinduced),isaglycosylatedreceptortyrosinekinasethatplaysacentralroleinepithelialmorphogenesisandcancerdevelopment.HGFRissynthesizedasasinglechainprecursorwhichundergoescotranslationalproteolyticcleavage.ThisgeneratesamatureHGFRthatisadisulfide-linkeddimercomposedofa50kDaextracellularalphachainanda145kDatransmembraNEBetachain(1,2).Theextracellulardomain(ECD)containsasevenbladedbeta-propellersemadomain,acysteine-richPSI/MRS,andfourIg-likeE-setdomains,whilethecytoplasmicregionincludesthetyrosinekinasedomain(3,4).ProteolysisandalternatesplicinggenerateadditionalformsofhumanHGFRwhicheitherlackofthekinasedomain,consistofsecretedextracellulardomains,oraredeficientinproteolyticseparationofthealphaandbetachains(5-7).Thesemadomain,whichisformedbyboththealphaandbetachainsofHGFR,mediatesbothligandbindingandreceptordimerization(3,8).Ligand-inducedtyrosinephosphorylationinthecytoplasmicregionactivatesthekinasedomainandprovidesdockingsitesformultipleSH2-containingmolecules(9,10).HGFstimulationinducesHGFRdownregulationviainternalizationandproteasome-dependentdegradation(11).Intheabsenceofligand,HGFRformsnoncovalentcomplexeswithavarietyofmembraneproteinsincludingCD44v6,CD151,EGFR,Fas,Integrinalpha6/beta4,PlexinsB1,2,3,andMSPR/Ron(12-19).Ligationofonecomplexcomponenttriggersactivationoftheother,followedbycooperativesignalingeffects(12-19).Formationofsomeoftheseheteromericcomplexesisarequirementforepithelialcellmorphogenesisandtumorcellinvasion(12,16,17).ParacrineinductionofepithelialcellscatteringandbranchingtubulogenesisresultsfromthestimulationofHGFRonundifferentiatedepitheliumbyHGFreleasedfromneighboringmesenchymalcells(20).Geneticpolymorphisms,chromosomaltranslocation,overexpression,andadditionalsplicingandproteolyticcleavageofHGFRhavebeendescribedinawiderangeofcancers(1).WithintheECD,humanHGFRshares86%-88%aasequenceidentitywithcanine,mouse,andratHGFR.
