ErbB3,alsocalledHer3(humanepidermalgrowthfactorreceptor3)inhumans,isatypeItransmembraneglycoproteinthatisamemberoftheErbBfamilyoftyrosinekinasereceptorsnamedforaviraloncogene(1‑3).ErbBfamilymembersserveasreceptorsfortheEGFfamilyofgrowthfactors(1‑3).MouseErbB3containsa19aminoacid(aa)signalsequence,a622 aaextracellulardomain(ECD),a24 aatransmembraneregion,anda677aacytoplasmicdomain(4).HumanErbB3hasfourisoformscreatedbyintronread-throughandtruncationofthemolecule(5).ThreeofthesearesecretedandatleastonecaninhibitErbB3activity(6).LittleinformationisavailableconcerningmouseErbB3isoforms.ThemouseErbB3ECDshares97%,93%,92%,91%,89%and88%aaidentitywithrat,human,bovine,equine, canineandopossumErbB3,respectively.ErbB3isfoundinepithelialcelllayersofgastrointestinal,reproductive,urinary,endocrineandnervoussystems,skinandmuscle(3).AmongErbBfamilymembers,onlyErbB3lacksaworkingkinasedomain,requiringheterodimerizationwithanotherErbBreceptorforsignaling(1‑3).TheheterodimerofErbB3withErbB2,whichhasnoknownligandsofitsown,isexpressedinthemajorityofbreast,skin,ovaryandgastrointestinaltumorsandtransducesahighlymitogenicsignalinresponsetoneuregulin1(NRG1;heuregulin1)orNRG2(3, 7‑9).TheseligandsalsobindErbB4(1).SignalingisaidedbythesixconsensusbindingmotifsfortheSH2domainandonefortheSH3domainoftheregulatoryp85subunitofphosphoinositide3-kinase(10, 11).Deletionstudiesinmicedemonstratenon-redundantrolesforErbB3indevelopmentofSchwanncells,neuralcrestcellsandheartvalves(12, 13).ErbB3,ErbB2andneuregulinareallrequiredforformationofthesympatheticnervoussystem(14).
