Thegalectinsconstitutealargefamilyofcarbohydrate-bindingproteinswithspecificityforN-acetyl-lactosamine-containingglycoproteins.Atleast14mammaliangalectins,whichsharestructuralsimilaritiesintheircarbohydraterecognitiondomains(CRD),havebeenidentifiedtodate.Thegalectinshavebeenclassifiedintotheprototypegalectins(-1,-2,-5,-7,-10,-11,-13,-14),whichcontainoneCRDandexisteitherasamonomeroranoncovalenthomodimer;thechimeragalectins(galectin-3)containingoneCRDlinkedtoanonlectindomain;andthetandem-repeatgalectins(-4,-6,-8,-9,-12)consistingoftwoCRDsjoinedbyalinkerpeptide.Galectinslackaclassicalsignalpeptideandcanbelocalizedtothecytosoliccompartmentswheretheyhaveintracellularfunctions.However,viaoneormoreasyetunidentifiednon-classicalsecretorypathways,galectinscanalsobesecretedtofunctionextracellularly.Individualmembersofthegalectinfamilyhavedifferenttissuedistributionprofilesandexhibitsubtledifferencesintheircarbohydrate-bindingspecificities.Eachfamilymembermaypreferentiallybindtoauniquesubsetofcell-surfaceglycoproteins.
