Thesystemusesabiotinylatedoligo(dT)primertohybridizeathighefficiencyinsolutiontothe3´poly(A)regionpresentinmostmatureeukaryoticmRNAspecies.Thehybridsarecapturedandwashedathighstringencyusingstreptavidincoupledtoparamagneticparticlesandamagneticseparationstand.ThemRNAiselutedfromthesolidphasebythesimpleadditionofribonuclease-freedeionizedwater.ThisprocedureyieldsanessentiallypurefractionofmaturemRNAafteronlyasingleroundofmagneticseparation.
