HepatitisBisasmallDNAvirusthatcontainsonly4openreADIngframesinitsgenome.ThreeoftheseORFshavebeenidentifiedastheenvelope,capsidandpolymerasegenes,whilethefunctionofthefourthhasremainedunknownuntilrecently.ThefourthreadingframehasbeencalledtheXgeneandproducesHBxproteinthatisessentialforviralreplicationinlivercellsandactsbroadlytoactivateavarietyoftranscriptionfactors.OnehypothesisofHBxactivityhasbeenthatitentersthenucleustoactasatranscriptionalregulator.OnepieceofevidencesupportingthismechanismofHBxactionisthatHBxinteractswiththetranscriptionfactorCREBandincreasesitsDNA-bindingactivity.ThismechanismdoesnotexplainalloftheeffectsofHBxhowever.AnotherhypothesisfortheHBxmechanismofactionisthatHBxincreasescalciumreleaseintothecytoplasm.ThismechanismofactionisconsistentwiththevarietyoftranscriptionfactorsandkinasesaffectedbyHBxsincecalciumsignalingaffectsmanydifferentsignalingpathways.HBxdoesnotappeartointeractdirectlywithRasorproteinkinasesbutdoesappeartoactivatethecalciumactivateproteinkinasePyk2.AdominantnegativeformofPyk2blocksHBxactivityandtheinvolvementofcalciuminHBxactivationofPyk2isindicatedbytheactionofseveralMolecularprobesthataltercytosoliccalciumsignaling.BAPTA-AMchelatescytosolicandblocksHBxaction.CyclosporinAblocksmitochondrialcalciumsignalingandalsoblockedHBxsignaling.CGP37157isacompoundthatblocksthemitochondrialsodiumcalciumpumpandalsoblockshepatitisBviralreplication,indicatingthatmitochondrialcalciumsignalingisthetargetofHBxfunction.Compoundsthatelevatecytoplasmiccalciumlevels(valinomycinandthapsigargin)increasetherateofreplicationofvirusesthatlackHBx,furthersupportingtheroleofHBxasaninducerofintracellularcalciumsignaling.ThemechanismbywhichHBxcausescalciumreleaseisnotyetknown.SrcactivationappearstoinduceviralreplicationwhileRassignalingdoesnot,suggestingthattheactivationofRassignalingbyHBxmayplayadistinctfunction,perhapstheinductionofcancer.
