Addgene has measured this occurs in 0.01-0.03% of viral vectors in our typical production protocol. This can lead to a small number of cells exhibiting Cre-independent transgene expression in vivo. To address this, we recommend titrating to find the optimal AAV dosage required for Cre-dependent transgene expression and function in vivo. 紫外-可见分光光度计由5个部件组成:①辐射源。必须具有稳定的、有足够输出功率的、能提供仪器使用波段的连续光谱,如钨灯、卤钨灯(波长范围350~2500纳米),氘灯或氢灯(180~460纳米),或可调谐染料激光光源等。②单色器[1]。它由入射、出射狭缝、透镜系统和色散元件(棱镜或光栅)组成,是用以产生高纯度单色光束的装置,其功能包括将光源产生的复合光分解为单色光和分出所需的单色光束。③试样容器,又称吸收池。供盛放试液进行吸光度测量之用,分为石英池和玻璃池两种,前者适用于紫外到可见区,后者只适用于可见区。容器的光程一般为0.5~10厘米。④检测器,又称光电转换器。常用的有光电管或光电倍增管,后者较前者更灵敏,特别适用于检测较弱的辐射。近年来还使用光导摄像管或光电二极管矩阵作检测器,具有快速扫描的特点。⑤显示装置。这部分装置发展较快。较高级的光度计,常备有微处理机、荧光屏显示和记录仪等,可将图谱、数据和操作条件都显示出来。
仪器类型则有:单波长单光束直读式分光光度计,单波长双光束自动记录式分光光度计和双波长双光束分光光度计。
应用范围包括:①定量分析,广泛用于各种物料中微量、超微量和常量的无机和有机物质的测定。②定性和结构分析,紫外吸收光谱还可用于推断空间阻碍效应、氢键的强度、互变异构、几何异构现象等。③反应动力学研究,即研究反应物浓度随时间而变化的函数关系,测定反应速度和反应级数,探讨反应机理。④研究溶液平衡,如测定络合物的组成,稳定常数、酸碱离解常数等。
荧光分析法 利用某些物质被紫外光照射后所发生的能反映出该物质特性的荧光,
可以进行定性或定量分析的方法。

荧光增白剂其实是一种荧光染料,或者说是种白色染料,它的特性是能激发入射光线产生荧光,使所染物质获得类似荧石的闪闪发光的效应,使肉眼看到的物质很白,达到增白的效果。
从国家质监局网站了解到,纺织、造纸、洗涤剂等产品中,国家是允许使用荧光增白剂,但荧光剂的使用并非没有禁忌,我国食品卫生法第六条明确规定,食品、食品包装、餐巾纸禁止使用荧光增白剂。
荧光剂对人体的危害目前国内还没有定论,也没有具体证据证明荧光剂吸收多少才会对人体造成伤害,但是在国外,荧光增白剂确实被列为潜在致癌物质之一。

所谓审时度势,关键在于要搞清楚基金究竟要赞助那些人,一般来说,国家设立基金,目的就是赞助那些能够在科研上有突破的人
。判断那些人能够在科研上有突破,这个就依靠评委来判断了,要知道国基从来不会雪中送炭,只会锦上添花。
你要申请,首先要想,假如你是评委,你写的这封标书送到你手上了,让你来判断是不是会有突破性成果,你怎样来判断?
你肯定说看工作基础,对了,这几乎是肯定的。就是一个申请青年基金的人,尽管不是很强调工作基础,但你想,现在评委的手里绝对不是就你一个基金标书,现在哪个角落、那个地方,可以说只要是人都在想办法申请基金,标书多的吓死人。评委不可能看到是青年基金,就不看工作基础了。所以不要认为申请青年基金,就可以不在工作基础上下功夫了。
那么,问题来了,如何在工作基础的书写上下功夫?或者说如何让评委一看就知道你是有工作基础的人?
当然是你发表的文章了,如果你在nature上发表一篇文章,兄弟,你不光今年可以中,明年也有你的份,而且,你申请20万,人家会说20万怎么会够?给40万吧。
一般来说,只要你能够在你从事的领域10%最高影响力上发表过一两篇文章,在工作基础方面你就不用愁了。现在的国内学者非常看重SCI的影响因子,因此呢,是人就知道这说明你在科研方面能够有突破。
因此工作基础最重要的是你有牛B文章,而不在于你发表文章的多少。从这个意义上来说,很多中文文章都是~~,呵呵,那个词太难听,不说了。
因此呀,奔2007国基的兄弟们,第一件要做的事情就是埋头工作,搞上几年,来一篇牛文。这是最重要的。
如果没有牛文,就拉那些有牛文的人入股,呵呵,这也应该算工作基础。有人说了,我真的没有,但还想申请,哪怎么办?兄弟不要急,请看第二计,瞒天过海。
