USAGE of the OVA Conjugated Myostatin (MSTN)
Reconstitute in PBS (pH7.4) to a concentration of 0.1-1.0 mg/mL. Do not vortex.
STORAGE of the OVA Conjugated Myostatin (MSTN)
Avoid repeated freeze/thaw cycles. Store at 2-8°C for one month. Aliquot and store at -80°C for 12 months.
STABILITY of the OVA Conjugated Myostatin (MSTN)
The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

我现在做关于细胞膜分子交联,然后用western检测多聚体的实验
遇到很大的困难
刚开始用真核细胞做,就发现细胞交联后提取蛋白,比未交联的细胞蛋白总量减少
后来用原核先提取蛋白做实验
发现交联终止后,做western交联的孔曝光没有条带,后来用同样的方法重新做了一次PAGE,染胶,发现蛋白不见了
为什么交联后蛋白减少或是不见了呢
我用的都是Pierce公司的交联剂,用过EGS和BOSCOES,都有同样的问题
方法是综合说明书和一片文献上来的
到底问题出在哪里呢
请各位大侠帮帮忙

指在光、热、高能辐射、机械力、超声波和交联剂等作用下,大分子链间通过化学键联结起来,形成网状或体形结构高分子的过程。橡胶的硫化、不饱和树脂的交联、环氧树脂的熟化等都是化学交联的例子。通过化学交联可改善聚合物的性能。如聚乙烯的化学交联可提高其强度和耐热性,又如皮革的鞣制过程是利用其蛋白质分子与甲醛作用,生成交联桥,以至失去溶解性。
明胶与干酪素都是蛋白质类,其分子结构同属氨基酸,但前者比后者价格便宜很多,而且在我国的产盆很大,因此具有代替干酪素做涂料胶粘剂的可能性。
在许多的细胞生命活动中,例如DNA复制、mRNA转录与修饰以及病毒的感染等都涉及到DNA与蛋白质之间的相互作用的问题.
重组DNA技术的发展,人们已分离到了许多重要的基因.现在的关键问题是需要揭示环境因子及发育信号究竟是如何控制基因的转录活性.为此需要:
a、鉴定分析参与基因表达调控的DNA元件;
b、分离并鉴定这些顺式元件特异性结合的蛋白质因子;
这些问题的研究都涉及到DNA与蛋白质之间的相互作用.
研究DNA-蛋白质相互作用的实验方法主要包括:

c、甲基化干扰实验; d、体内足迹实验; f、拉下实验.
