INTENDEDUSE
ACTICHROME®Heparin(anti-FXa)isachromogenicassayintendedforthequantitativedeterminationofunfractionatedandlowmolecularweightheparinsinhumanplasmabymeasurementoffactorXainhibition.
PRINCIPLE
TheinhibitoryeffectofantithrombinIII(AT-III)onthrombin,factorXaandothercoagulationserineproteasesinplasmaisincreasedseveralthousandfoldbyheparin.Thisinhibitionaccountsfortheanticoagulanteffectofheparin.Thequantitativedeterminationofplasmaheparinlevelsbythe
measurementoftheiranti-Xaactivityisanecessarytoolformonitoringtreatmentefficacy.
Lowmolecularweightheparin(LMWH)preparationsappeartocatalyzethereactionbetweenfactorXaandAT-IIImorereadilythanthereactionbetweenthrombin(FIIa)andAT-III.Unfractionatedheparin(UFH)catalyzesbothreactionsequally.ThefactorXainhibitiontestisthemostusefulassay
coveringthewidestvarietyofheparinpreparations.Intheassay,therateoffactorXainhibitionisdirectlyproportionaltotheheparinconcentrationsincebothfactorXaandAT-IIIareinexcess.TheresidualfactorXaactivityisinverselyproportionaltotheheparinconcentration.1,2
