2.GeneralInformation Theinfluenzainfectionisanacutefeverishvirusinfection,whichprincipallyleadstoanillnessof therespiratorytractandappearsasanepidemicorpandemic.Theinfectionmostlyresultsfroma dropletinfection.Thevirusspreadsfromthemucousmembraneoftheupperrespiratorytothe wholebronchialtract.Therethevirusanditstoxincanleadtoaseriousinflammationofthe bronchialmucosaandadamageofthevessels. Afteranincubationtimeof1to3daysthe symptomsappearsuddenly:Followedbyafast increaseoftemperature,oftenaccompaniedby shivering,thecatarrhalleADIngsymptomappears,whichcontributetothecoursebesidepainfuldry cough,tracheitis,laryngitisandfrequentlyarhinitisandconjunctivitis. TheInfluenzavirusesformavirusgroupwith principallysimilarmorphological,chemicaland BIOLOGicalfeatures.ThetypesA,BandCweredefined,fromwhichmanyothervariantsareknown. Thedistinctionofthetypeswillbepossiblebythedifferentantigenicityoftheirnucleoproteins, whicharecoatedbyamatrixproteinwithtype-specificantigenicity,too.However,bothinternal antigensareoflessimportancefortheimmunity.Theessentialantigensarethehemagglutininand theneuraminidase.Botharesurfaceantigensandsubjecttoapermanentchangeoftheir antigenicity,whichiscalleddriftorshift.TheappearenceofpermanentnewInfluenzaepidemics andpandemicsareparticularlyfacilitatedbyanantigenvariABIlity,becausethenewdriftorshift variantsinfectapopulationwhichisonlypartlyimmuneorinanextremecasecompletely susceptibletothedisease. 3.PrincipleoftheTest TheGenWayInfluenzaBIgMantibodytestkit isbasedontheprincipleoftheenzyme immunoassay(EIA).InfluenzaBantigenisboundonthesurfaceofthemicrotiterstrips.Diluted sampleserumorready-to-usestandardsarePipettedintothewellsofthemicrotiterplate.Abinding betweentheIgMantibodiesoftheserumandthe immobilizedInfluenzaBantigentakesplace. Afteraonehourincubationatroomtemperature,theplateisrinsedwithdilutedwashsolution,in ordertoremoveunboundmaterial.Thenready-to-useanti-human-IgMperoxidaseconjugateis addedandincubatedfor30minutes.Afterafurtherwashingstep,thesubstrate(TMB)solutionis pipettedandincubatedfor20minutes,inducingthedevelopmentofabluedyeinthewells.The colordevelopmentisterminatedbytheadditionofastopsolution,whichchangesthecolorfrom bluetoyellow.Theresultingdyeismeasuredspectrophotometricallyatthewavelengthof450nm. TheconcentrationoftheIgMantibodiesisdirectlyproportionaltotheintensityofthecolor.
