- 400nmolesoffattyacidstandard(pentadecanoicacidsC15)
METHODS:- Prepare1MMeOH-HClreagentaccordingtotheinstructiongivenwiththekit.
- Addinternalstandard(C15fattyacid)togiveafinalconcentrationof400nmoles/100ml(usually10mgofC15in100mlofreagent.
- Weigh10mgofwholecellsor1mgofLPSintoacleanscrew-captube.
- Add1mlofMeOH-HClreagent/internalstandardintoeachtubeandvortex.
- Heatat100oCfor20minutes.(Note:eachtubeshouldbeverywellsealedwithteflontapeandgreasetopreventevaporation.)
- Sonicateandheatat100oCovernight.
- Neutralizeaciditywith0.5NNaOH.TestpHwithpHpaper.
- Centrifugeinclinicalcentrifugefor5minutes.Savesupernatentincleanglassvial.
- DogaschomatographywithprogrammedREWHmethod.
