Procedure
- Dissolve10mgofcommercialRNAin250ml.ofslightlyalkalinedistilledwater.Useavolumetricflaskandproperanalyticaltechnique.Thiswillgiveastandardsolutionof40microgramsRNA/ml.
- Prepareaseriesofdilutionssothatyouhave40,20,10,5and2.5microgramsofRNAperml.
- TurnonaUVspectrophotometerandadjustthewavelengthto260nm.Usethealkalinewatertoblankthespectrophotometerat260nm.
- ReadtheA
ofeachofthestandards.PlottheAvstheconcentrationofRNAandcalculatetheextinctioncoefficient. - Dissolveyourisolated,precipitatedRNAin10.0mlofalkalinewater.Prepareaserialdilutionfor1/10,1/100,1/1000and1/10000.Measuretheabsorbanceofeachat260nmand,usingthedilutionwhichgivesareADIngbetween.1and1.5absorbanceunits,computetheconcentrationofRNAinyoursample.
