Themechanismofimmunetoleranceistobefurtherunderstood.ThepresentstudyaimstoinvestigatetheroleoftheCardiacendothelialcell(CEC)-derivedexosomesintheinductionofregulatoryBcells.Inthisstudy,CECswereisolatedfromthemouseheart.Exosomeswerepurifiedfromtheculturesupernatantoftheprimaryendothelialcells.ThesuppressorfunctionsoftheregulatoryBcellsweredeterminedbyflowcytometry.TheresultsshowedthattheCEC-derivedexosomescarriedintegrinv6.Exposuretolipopolysaccharide(LPS)inducedBcellstoexpressthelatenttransforminggrowthfactor(TGF)-,thelatterwasconvertedtotheactiveform,TGF-,bytheexosome-derivedv6.TheBcellsreleasedTGF-inresponsetore-exposuretotheexosomesintheculture,whichsuppressedtheeffectorTcellproliferation.WeconcludethatCEC-derivedexosomeshavethecapacitytoinduceBcellswithimmunesuppressorfunctions.
