相关技术背景
microRNA(简称miRNA)是一种细胞内源性的小ncRNA(Non-codingRNA,非编码RNA),miRNA网络被发现是基因表达调控的重要层次,除了参与生长、发育、分化、凋亡等重要生物学过程,还与疾病的发生、发展、转归关系密切。
本项技术和产品的工作原理
使用病毒载体携带的miRNA的传感器,制备出包括了全部miRNA传感器种类的病毒载体,并将其包被在细胞培养孔板上,采用病毒反向感染法使包被在孔板上的病毒载体反向感染后加入的细胞,以用于miRNA传感器检测细胞的miRNA活性。
由于使用可分泌表达的、可受细胞内miRNA活性调节的报告基因Gluc做为指示待检测指标的生物传感器,因此,可以通过检测细胞培养上清中的Gluc表达水平来体现细胞内miRNA的适时的活性,并可多次反复取上清进行检测。
Gluc的工作原理
Gaussia荧光素酶(Gluc,Gaussialuciferase),其酶促反应底物为:腔肠素(coelenterazine)
发光反应:
在Gluc催化下,发生下述反应并产生荧光:
Coelenterazine+O2→coelenteramide+CO2
Fluc的工作原理
萤火虫荧光素酶
(Fluc,fireflyluciferase),
其酶促反应底物为:D-luciferin

本产品的应用范围
1、细胞miRNA活性谱的检测,产生细胞指纹,建立细胞特征信息库;
2、miRNA将成为疾病筛查、诊断、疗效评价和预后判断上一种新的标志物;
3、用于肿瘤检测和治疗评价、机理研究;
4、用于新药研发过程中的大量的药物筛选工作,药敏、药靶研究开发;

本产品的特点
本产品可实时、连续、动态检测活细胞内miRNA活性谱的变化。
该产品检测的是活细胞的miRNA活性而不是miRNA的拷贝数,这与其它方法不同。

实验流程及步骤
1、将等量单细胞悬液按10(e)4-5个细胞/孔加入到miRNAA-Sensor板(96孔细胞培养板)中,置于CO2培养箱培养;
2、用96孔荧光检测仪测细胞上清的荧光强度,获得原始数据。
miRNA活性单位为RIF(RelativeInhibitedFold,相对抑制倍数)。
组织块的miRNA活性检测:手术、活检取样获得的无污染的、有活性的组织块,经消化、处理成单细胞悬液,进行原代细胞培养,达到上述步骤1中所描述的细胞数量,即可按上述步骤进行miRNA活性检测。
实验的硬件条件
1、可进行动物细胞培养的实验室,主要包括:超净工作台、CO2培养箱;
2、96孔荧光检测仪:miRNA活性检测结果为全自动、机检格式、定量数据。
以上为使用本产品所需基本条件。
3、如需大量药物筛查测细胞的miRNA活性变化,则可使用叠板系统与96通道自动移液工作站组成全自动工作站,进行细胞的加样、加药物、吸取上清等操作。
