Astissuesgrowtheyrequireangiogenesistooccuriftheyaretobesuppliedwithbloodvesselsandsurvive.Factorsthatinhibitangiogenesismightactascancertherapeuticsbyblockingvesselformationintumorsandstarvingcancercells.Thrombospondin-1(TSP-1)isaproteinthatinhibitsangiogenesisandslowstumorgrowth,apparentlybyinducingapoptosisofmicrovascularendothelialcellsthatlinebloodvessels.TSP-1appearstoproducethisresponsebyactivatingasignalingpathwaythatbeginswithitsreceptorCD36atthecellsurfaceofthemicrovascularendothelialcell.Thenon-receptortyrosinekinasefynisactivatedbyTSP-1throughCD36,activatingtheapoptosisinducingproteaseslikecaspase-3andp38proteinkinases.p38isamitogen-activatedkinasethatalsoinducesapoptosisinsomeconditions,perhapsthroughAP-1activationandtheactivationofgenesthatleadtoapoptosis.
