2.GeneralInformation
Theinfluenzainfectionisanacutefeverishvirusinfection,whichprincipallyleadstoanillnessof therespiratorytractandappearsasanepidemicorpandemic.Theinfectionmostlyresultsfroma dropletinfection.Thevirusspreadsfromthemucousmembraneoftheupperrespiratorytothe wholebronchialtract.Therethevirusanditstoxincanleadtoaseriousinflammationofthe bronchialmucosaandadamageofthevessels. Afteranincubationtimeof1to3daysthe symptomsappearsuddenly:Followedbyafast increaseoftemperature,oftenaccompaniedby shivering,thecatarrhalleADIngsymptomappears, whichcontributetotheclinicalcoursebeside painfuldrycough,tracheitis,laryngitisandfrequentlyarhinitisandconjunctivitis. TheInfluenzavirusesformavirusgroupwith principallysimilarmorphological,chemicaland BIOLOGicalfeatures.ThetypesA,BandCweredefined,fromwhichmanyothervariantsareknown. Thedistinctionofthetypeswillbepossiblebythedifferentantigenicityoftheirnucleoproteins, whicharecoatedbyamatrixproteinwithtype-specificantigenicity,too.However,bothinternal antigensareoflessimportancefortheimmunity.Theessentialantigensarethehemagglutininand theneuraminidase.Botharesurfaceantigensandsubjecttoapermanentchangeoftheir antigenicity,whichiscalleddriftorshift.TheappearenceofpermanentnewInfluenzaepidemics andpandemicsareparticularlyfacilitatedbyanantigenvariABIlity,becausethenewdriftorshift variantsinfectapopulationwhichisonlypartlyimmuneorinanextremecasecompletely susceptibletothedisease. ThedeterminationoftheInfluenzatype(A,B,andC)givesboththeclinicianandepidemiologist importantindicationsforfurtheractions.ThusInfluenzaBoftenleadstoaseriousclinicalcourse andanepidemicspreadofthevirus.Similarly,duringanInfluenzaAepidemic,theepidemiological importanceandderivedmeasuresfortheprotectionoftheindividualandpopulationprimarilystand intheforegroundtogetherwiththeseverityoftheclinicalsymptoms.
3.PrincipleoftheTest
TheGenWayInfluenzaAIgAantibodytestkitisbasedontheprincipleoftheenzyme immunoassay(EIA).InfluenzaAantigenisboundonthesurfaceofthemicrotiterstrips.Diluted patientserumorready-to-usestandardsarePipettedintothewellsofthemicrotiterplate.Abinding betweentheIgAantibodiesoftheserumandthe immobilizedInfluenzaAantigentakesplace. Afteraonehourincubationatroomtemperature,theplateisrinsedwithdilutedwashsolution,in ordertoremoveunboundmaterial.Thenready-to-useanti-human-IgAperoxidaseconjugateis addedandincubatedfor30minutes.Afterafurtherwashingstep,thesubstrate(TMB)solutionis pipettedandincubatedfor20minutes,inducingthedevelopmentofabluedyeinthewells.The colordevelopmentisterminatedbytheadditionofastopsolution,whichchangesthecolorfrom bluetoyellow.Theresultingdyeismeasuredspectrophotometricallyatthewavelengthof450nm. TheconcentrationoftheIgAantibodiesisdirectlyproportionaltotheintensityofthecolor.
