DigestionofBacPAK6DNAatthreesiteswithBsu36Iremoves lacZ andpartofORF1629,whichisanessentialAcMNPVgene. ThelinearvirusDNAisunabletoinitiateareplicationcycleininsectcells.
Co-transfectionofinsectcellswithlinearBacPAK6DNAandacompatIBLetransfervectorcontainingthecompleteORF1629andaforeigngeneofinterestresultsinrecombinationandrestorationofthecircular,infectiousvirusgenome. Over95%ofthevirusderivedfromtheco-transfectioncomprisesrecombinantvirus. Usually,asingleplaqueassaytitrationincubatedwithX-galandcounterstainedwithneutralredissufficienttodifferentiateparentalbluefromrecombinantwhite(colourless)plaques. ThesecanbereADIlyisolatedandamplifiedtoworkingstocksofpurifiedrecombinantvirusinafewdays.
