UsingtheflashBAC™systemiseasierandquickerthanotherbaculovirusexpressionsystemsbecausethereisnoneedtoseparaterecombinantvirusfromparentalvirusbyplaque-purificationoranyothermeans;onlyrecombinantvirusisproducedaftertheco-transfection. Becausetheproductionofrecombinantvirushasbeenreducedtoaonestepprocedureininsectcells,itamenabletohighthroughputandautomatedproductionsystems. However,itisalsoofbenefittothesmallresearchgroupjustrequiringoneorafewrecombinantbaculovirusespreparedinindividualdishesofcells.
Baculovirusgenomescontainseveralauxillarygenes,whicharenon-essentialforreplicationininsectcellculture.Oneoftheseischitinase(chiA),whichencodesanenzymewithexo-andendochitinaseactivity.Inaninfectedinsect,chitinasetogetherwiththecysteineproteasecathepsin,facilitateshostcuticlebreakdownandtissueliquefactionattheverylatestagesofinfection,soreleasingthevirustoinfectmorehosts.ConfocalandelectronmicroscopyobservationsofinsectcellsinfectedwithAcMNPVhaveshownthatchitinaseistargetedtotheendoplasmicreticulumwhereitisdenselypackedinaparacrystallinearray,severelycompromisingthefunctionandefficacyofthesecretorypathway.DeletionofchiAfromflashBAC™hasimprovedtheefficacyofthesecretorypathwayandresultedinagreatlyenhancedyieldofsecretedandmembranetargetedproteins. Deletionofv-cathreducesthechancesoftherecombinantproteinbeingdegraded.
