InVitro

Puromycinblocksproteinsynthesisafteraminoacyl-sRNAformation,andatthesametimeitleadstotheaccumulationofsmallpeptides.Bothoftheseeffectsappeartobeduetothesplittingofribosome-boundpeptidyl-sRNA,4whichresultsinreleaseofincompletepeptidechains.[1].Puromycin,ananalogofthe3"endofaminoacyl-tRNA,causesprematureterminationoftranslationbybeinglinkednon-specificallytogrowingpolypeptidechains.Puromycinhastwomodesofinhibitoryaction.Thefirstisbyactingasanacceptorsubstratewhichattackspeptidyl-tRNAinthePsitetoformanascentpeptide.Thesecondisbycompetingwithaminoacyl-tRNAforbindingtotheA"site[2].Whenusedinminimalamounts,puromycinincorporationinneosynthesizedproteinsreflectsdirectlytherateofmRNAtranslationinvitro.PuromycinimmunodetectionisanadvantageousalternativetorADIoactiveaminoacidlabeling.Itallowsthedirectevaluationoftranslationactivityinsinglecellsbyimmunofluorescencemicroscopyandinheterogenouspopulationsofcellsbyfluorescenceactivatedcellsorting[3].
