Description

MPTP(hydrochloride)isaprodrugtotheneurotoxinMPPwhichcausespermanentsymptomsofParkinson"sdisease.

InVitro

Pretreatmentwith50mM4-phenylpyridine,reducesIC50(concentrationfor50%inhibitionoftwitchamplitude)valuesofMPTPfrom53to18mMandd-tubocurarinefrom0.7to0.3mM,respectively,inmousephrenicnerve-diaphragm[3].

InVivo

ThetoxiceffectofMPTPcanbecompletelyabolishedinvivobytreatmentwithamonoamineoxidaseinhibitorandpotentiatedbyaninhibitorofcatechol-O-methyltransferase[1].AllopurinolpotentiatedtheMPTP(35mg/kg)-induceddecreaseintheDOPAC+HVA/DAratioandincreaseinstriatalAAoxidationoftherat[2].Gas1expressionsaresignificantlyelevatedinthemajorityofthereactiveastrocytesofthebrainswithLPSorMPTPinsultsinanimalmodels[4].

MPTPisformulatedin0.9%saline.

ForthepreparationoftheLPSratmodelandtheMPTPmousemodel,thetreatmentsoftheanimalsareperformed.Briefly,adultratsreceiveunilateralinjectionsofLPS(0.5 μLof10 μg/μLdilutedin0.9%saline)intothemedialforebrainbundle(MFB)atthefollowingcoordinates,AP-4.2 mm,L1.5 mm,andV7.8 mm,andintothecontralateralsidewiththesamevolumeof0.9%saline.AdultmiceareadministeredintraperitonealinjectionsofMPTPof25 mg/kgperdayforfivecontinuousdays,andthesamevolumeofsalineisinjectedasacontrol.Alltheanimalsaresacrificedatweek1,2,3,or4aftertheLPSorMPTPinjections.Thebrainsamplesarecollectedforthesubsequentimmunohistochemistryandwesternblotexperiments.MCEhasnotindependentlyconfirmedtheaccuracyofthesemethods.Theyareforreferenceonly.
