ShortDescription:InthisassaytheHaptoglobinpresentinsamplesreactswiththeanti-Haptoglobinantibodieswhichhavebeenadsorbedtothesurfaceofpolystyrenemicrotitrewells.Aftertheremovalofunboundproteinsbywashing,anti-HpantibodiesconjugatedwithhorserADIshperoxidase(HRP),areadded.Theseenzyme-labeledantibodiesformcomplexeswiththepreviouslyboundHp.Followinganotherwashingstep,theenzymeboundtotheimmunosorbentisassayedbytheadditionofachromogenicsubstrate,3,3’,5,5’-tetramethylbenzidine(TMB).ThequantityofboundenzymevariesdirectlywiththeconcentrationofHpinthesampletested;thus,theabsorbance,at450nm,isameasureoftheconcentrationofHpinthetestsample.ThequantityofHpinthetestsamplecanbeinterpolatedfromthestandardcurveconstructedfromthestandards,andcorrectedforsampledilution.AdditionalInformation:
